  • Essay / Effects of Surfactants on Suppository Absorption

Suppositories are the frequently used dosage forms prescribed rectally. However, suppositories have a limited onset of action, which can be attributed to the poor drug release properties of the suppository base. Surfactants can increase drug release to facilitate absorption by reducing interfacial tension between rectal fluids and the suppository base. The objective of this study was to examine the effect of different concentrations of sodium lauryl sulfate and tween 80 on the drug release of paracetamol from palm kernel suppositories (HAMIN®). Sodium lauryl sulfate (SLS) and Tween 80 were used in this study to examine their effect on drug release from paracetamol suppositories prepared using HAMIN® base by the double pour method. The effect of SLS at concentrations of 0.25%, 0.5%, 0.75% and 1% w/w in suppositories was analyzed, the percentage increase in drug release was 26, respectively. .6%, 28.9%, 37.4% and 84.6%. But at the same concentrations, Tween 80 showed a higher drug release percentage than SLS, which was 0.79, 8.9, 10, and 10 times, respectively. It can be concluded that although SLS increased drug release from the formulated suppositories, it was not as effective as Tween 80.Keywords: Rectal route of administration, Suppository, paracetamol, Sodium lauryl sulfate, Tween 80, HAMIN® base.IntroductionThe oral route is the most common route of drug administration in daily practice. However, medications may not be suitable for oral administration in certain circumstances. Young children, the elderly, people experiencing nausea, vomiting, and unconscious patients have difficulty or are unable to swallow medications [1]. In these cases, the rectal route becomes a good al......middle of paper......auryl sulfate (SLS) will be used at different concentrations in palm kernel-based paracetamol suppositories (HAMIN ®) to examine its effect on drug release and to determine the most favorable concentration of SLS to increase the absorption rate of rectal paracetamol. Tween 80 is also known as polysorbate 80 or polyoxyethylene 20 sorbitan mono-oleate. [13] It is an oleate ester of sorbitol and its anhydrides. It is an oily liquid, clear or slightly opalescent, brownish yellow or colorless. Tween 80 is considered one of the most commonly used surfactants in suppository formulation. [14] It is a non-ionic surfactant widely used in the formulation of suppositories over the years, it is chosen to be compared with SLS on the drug release rate at the same concentrations, because such a study has not been previously reported with the HAMIN base®..
